Discovering High-Affinity Ligands for Proteins

This article reports on development of a method for producing high-affinity ligands in which small molecules that bind to proximal subsites of a protein are identified in an NMR-based screen and then linked together in their experimentally determined bound orientations. The method is called “SAR by NMR,” which stands for “structure-activity relationships by nuclear magnetic resonance.”
